Nanoparticles comprised of the poly(ethylene oxide)-b-poly (lactic acid) diblock copolymer (PEO-b-PLA) with and without the incorporation of lavender oil were prepared by nanoprecipitation. Diblock copolymers based on a fixed PEO block (5KDa) and two different PLA segments (4.5 or 10KDa) were used. The morphology, encapsulation efficiency, essential oil-polymer interaction and the release kinetics of the active agent in the nanoparticles, were evaluated. The hydrodynamic radius of the nanoparticles determined by light scattering was affected by the size of the poly(lactic acid) (PLA) block. The lavender essential oil encapsulation efficiency (at a concentration of 0.4 µL mL-1) determined by UV-VIS spectroscopy was in the range of 70-75%. The in vitro release suggests that the polymeric barrier is able to control the oil release.

Poli(acrilamida-co-dihexilacrilamida) (PAHM-0) e poli(acrilamida-co-acrilato de sódio-co-dihexilacrilamida) (PAHM-21) foram estudadas por espalhamento de raios-X a baixos ângulos (SAXS), espalhamento de luz (LS) e reologia. Os resultados de SAXS ressaltaram o caráter polieletrolítico da PAHM-21, com uma conformação altamente estendida em solução aquosa devido às repulsões entre as cargas, enquanto a PAHM-0 tem uma conformação em novelo aleatório. As medidas de LS indicaram que a PAHM-0 forma aglomerados intermoleculares em solução, na presença e na ausência de sal, mesmo com um conteúdo hidrofóbico menor que o descrito na literatura para poliacrilamidas associativas. Contudo, os resultados reológicos mostraram que, apesar de haver associação hidrofóbica, não há um aumento da viscosidade. Os resultados de LS da PAHM-21 sugerem que esse polímero forma, predominantemente, associações intramoleculares na presença de sais. Além disso, as medidas viscosimétricas mostram que a sua viscosidade diminui com a blindagem das cargas pela adição de sais.

Poly(acrylamide-co-dihexylacrylamide) (PAHM-0) and poly(acrylamide-co-sodium acrylate-co-dihexylacrylamide) (PAHM-21) were studied through small-angle X-ray scattering (SAXS), light scattering (LS) and rheology. SAXS results highlighted the polyelectrolyte character of PAHM-21, with highly extended conformation in aqueous solution owing to charge repulsion, while the PAHM-0 has a coil conformation. LS measurements indicated that PAHM-0 makes intermolecular clusters in solution, in presence and absence of salt, even with a lower hydrophobic content than that described in the literature to the associative polyacrylamides. However, the rheological results showed that there is not an enhancement of the viscosity although hydrophobic association takes place. LS results for PAHM-21 suggest that this polymer makes intramolecular associations mainly in the presence of salts. Furthermore, the viscosity measurements show that its viscosity decreases due to screening of the charges by the addition of salts.

Neste estudo, nanocarreadores lipídicos foram preparados a partir do triestearato de glicerila e óleo de rícino em diferentes proporções pela técnica de difusão do solvente a quente. Estearato de PEG-660 ou Poloxamer 188 e lecitina de soja foram usados como surfactante e cossurfactante, respectivamente. Características estruturais e físico-químicas das partículas foram investigadas por meio de espalhamento de luz dinâmico (DLS), calorimetria exploratória diferencial (DSC), espalhamento de raios X a altos ângulos (WAXS), microscopia de força atômica (AFM) e microscopia eletrônica de transmissão (TEM e cryo-TEM). Aanálise por DLS indicou que partículas menores foram obtidas quando o lipídio líquido foi usado sozinho para obter as nanoemulsões (NEs). Técnicas microscópicas (TEM e cryo-TEM) mostraram que as NEs apresentaram formas esféricas, enquanto as nanopartículas lipídicas sólidas (NLSs) e os carreadores lipídicos nanoestruturados (CLNs) apresentaram formas mais complexas. As NLSs exibiram estrutura cristalina lamelar, enquanto os CLNs exibiram uma estrutura mista composta de fase cristalina lamelar em contato com compartimento oleoso. Estes experimentos contribuem para um melhor entendimento da capacidade de incorporação e liberação de fármacos destes nanocarreadores.

In this study, drug delivery nanocarriers based on glyceryl tristearate and castor oil at different ratios were prepared by the hot solvent diffusion method. PEG 660-stearate or Poloxamer 188 and soybean lecithin were used as surfactant and co-surfactant, respectively. Structural and physicochemical characteristics of the particles were investigated by means of dynamic light scattering (DLS), differential scanning calorimetry (DSC), wide-angle X-ray scattering (WAXS), atomic force microscopy (AFM) and transmission electron microscopy (TEM and cryo-TEM). DLS data indicated that smaller particles were obtained when the liquid lipid was used alone to prepare nanoemulsions (NEs). TEM and cryo-TEM images showed that NEs presented spherical particles, whereas solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs) presented more complex shapes. SLNs exhibited lamellar crystal structure, while NLCs displayed a hybrid structure composed of a lamellar crystalline phase in contact with a liquid oil compartment. These experiments contribute to a better understanding of the structure and release performance of these drug carrier systems.

A engenharia de superfícies para o posicionamento de biomoléculas é uma etapa fundamental para a produção de biochips. Neste estudo, descreve-se uma nova estratégia para a preparação de nanodomínios contendo grupos ácido fosfônicos, os quais se ligam a uma família de proteínas (anexinas), de forma reversível em função da presença de íons cálcio. A abordagem desenvolvida consistiu no uso de precursores de poliestireno-b-poli(metacrilato de 2-hidroxietila) (PS-b-PHEMA) sintetizados por polimerização radicalar por transferência de átomos (ATRP) para a obtenção de cilindros de PS (fPS = 0.25-0.27) dispostos em morfologia hexagonal numa matriz de PHEMA. Os grupos hidroxila pendentes do PHEMA foram, em seguida, parcialmente (20%) convertidos em grupos ácido fosfônicos por reações sequenciais de fosforilação, sililação e metanólise. Este processo produziu derivados PS-b-P(HEMA-co-PEMA) que retêm a morfologia hexagonal, embora o grau de organização estrutural seja menor em relação aos precursores.

Surface engineering for precise positioning of biomolecules is of particular interest for controlled fabrication of biochips. In this study, a novel approach for the preparation of nanodomains with phosphonic diacid groups that were previously shown to reversibly bind to a family of proteins (annexins) in a calcium-dependent manner is described. The strategy makes use of polystyrene-b-poly(2-hydroxyethyl methacrylate) (PS-b-PHEMA) precursors prepared by atom transfer radical polymerization (ATRP) to obtain hexagonally packed PS cylinders (fPS = 0.25-0.27) in a PHEMA matrix. The pendant hydroxyl groups of PHEMA are then partially (ca. 20%) converted into phosphonic diacid groups via sequential reactions involving phosphorylation, silylation and methanolysis. This process produces PS-b-P(HEMA-co-PEMA) derivatives that still retain the hexagonal morphology, but their degree of structural organization is reduced comparatively to the precursors.

Uma poliacrilamida hidrofobicamente modificada e dois dos seus derivados parcialmente hidrolisados, contendo grupos hidrofóbicos e carboxila, foram preparados por polimerização micelar e pós-hidrólise. A massa molar, o segundo coeficiente do virial e o raio de giração foram determinados por espalhamento de luz estático (SLS). O espalhamento de luz dinâmico (DLS) e o espalhamento de raios-X a baixos ângulos (SAXS) foram utilizados respectivamente para determinar a formação de agregados e o tipo de empacotamento das cadeias em regime semi-diluído. O comportamento das soluções, em regime diluído e semi-diluído foi estudado por viscosimetria e reologia. A poliacrilamida modificada hidrofobicamente apresentou tendência à formação de agregados devido aos grupos hidrofóbicos, mas essa agregação não foi suficiente para aumentar a viscosidade aparente. Embora os derivados parcialmente hidrolisados não apresentassem a mesma tendência para agregação, eles apresentaram um comportamento anisotrópico devido à introdução de densidade de carga sobre a cadeia polimérica, a qual levou a uma conformação mais alongada da macromolécula e maior viscosidade.

A hydrophobically-modified polyacrylamide and two partially hydrolyzed derivatives containing hydrophobic and carboxylic groups were prepared by micellar polymerization and post-hydrolysis. The molecular weight, second virial coefficient and radius of gyration were determined by static light scattering (SLS). Dynamic light scattering (DLS) and small-angle X-ray scattering (SAXS) were employed to determine the aggregate formation and type of chain packing in the semidilute regime, respectively. The behavior of solutions in dilute and semidilute regimes was also studied by viscometry and rheology. The hydrophobically-modified polyacrylamide showed a tendency to form aggregates due to the hydrophobic groups, but not enough to increase apparent viscosity. The partially hydrolyzed derivatives did not show the same aggregate-forming tendency. Rather, they exhibited anisotropic behavior, due to the charge density introduced into the polymer chain, which led to a more elongated macromolecular conformation and higher viscosity.

Efeitos de adição de sal, concentração, presença de contraíons, temperatura e agente quelante em soluções aquosas de pectina, alginato de sódio e misturas destes foram analisadas por viscosimetria e espalhamento de luz dinâmico (DLS). A viscosidade intrínseca dos sistemas binários diminuiu com a adição de sal e com o aumento da temperatura, porém, mostrou-se insensível à adição de NaEDTA. Como esperado, a viscosidade intrínseca do sistema ternário alginato/pectina/água foi igual a média da viscosidade intrínseca determinada para os sistemas binários. Estudos de DLS indicaram a ocorrência de uma distribuição bimodal (presença dos modos de relaxação rápido e lento) para ambos os sistemas binários e ternários a 25 °C, refletindo um processo de agregação. No caso do modo lento de relaxação, um aumento significativo do raio hidrodinâmico foi observado tanto para os sistemas binários como ternário, na presença de NaCl e KCl a 25 °C. No entanto, a 80 °C, o raio hidrodinâmico correspondente ao modo lento em soluções de KCl foi praticamente constante para todos os sistemas estudados e não foi observado para o sistema binário alginato/solução aquosa.

The effects of added salt, concentration, counterions, temperature and chelating agent on aqueous solutions of pectin, sodium alginate and their mixtures were analyzed by viscosimetry and dynamic light scattering (DLS) techniques. The intrinsic viscosity of the binary systems decreased with the addition of salt and with temperature, while it was found to be insensitive to the addition of NaEDTA. As expected the intrinsic viscosity of the ternary alginate/pectin/water system was equal to the average of the intrinsic viscosity for the binary systems. The DLS studies indicated a bimodal distribution (fast and slow relaxation modes) for both binary and ternary systems at 25 °C, reflecting aggregation. A significant increase in the hydrodynamic radius, in the case of the slow mode, was observed for the binary and ternary systems in the presence of NaCl and KCl at 25 °C. However, at 80 °C the hydrodynamic radius for the slow mode in KCl solutions was practically constant for all the studied systems, except for the alginate binary solutions in which were not observed.

The effects of sorbitol and formaldehyde on the morphology, water absorption and mechanical properties of sodium alginate films were analyzed. The morphology of the films indicated the presence of small aggregates in the surface of uncrosslinked films, which disappeared with the crosslinking process. The water uptake and percentage of elongation increased with the addition of sorbitol in uncrosslinked films. At the same time, a decrease in tensile strength and Young's modulus occurred. The swelling ratio and water uptake of crosslinked alginate/sorbitol films decreased with an increase in sorbitol content suggesting an enhanced crosslinking density due to the presence of plasticizer.

A influência da estrutura química dos grupos mesogênicos e do tamanho dos grupos espaçadores, no comportamento de fase de uma série de cristais líquidos poliméricos de cadeia lateral (SCLCP), foram estudados utilizando-se espalhamento de raios-X a Baixo Ângulo (SAXS) e Microscopia Ótica de Luz Polarizada (POM). Análises do arranjo das mesofases em amostras não orientadas e orientadas por ação do campo magnético são descritas. O papel do tamanho do espaçador lateral no empacotamento local e na largura da camada esmética determinados nas mesofases SmA e SmC é elucidado. Os ângulos teta formados entre os grupos mesogênicos e a normal às camadas nas mesofases SmC foram determinados. Um estudo a respeito do grau de ordem em função da temperatura, para os polímeros esméticos foi possível através de medidas de SAXS. Uma ordenação particular em um dos SCLCPs estudados é relacionada com a coexistência de duas fases distintas.

The influence of the chemical structure of the mesogenic groups and the length of the spacer groups on the phase behavior in a series of side-chain liquid crystalline polyacrylates (SCLCP) have been studied using Small Angle X-ray Scattering (SAXS) and Polarized Optical Microscopy (POM). Analyses of the mesophase arrangement in unaligned and aligned samples by magnetic field are reported. The role of the spacer length on the local packing and on the thickness of the layers encountered in the SmA and SmC mesophases is elucidated. The tilt angles theta of the mesogenic cores related to the normal of the layers in the SmC mesophases are measured. A study about the degree of order as a function of temperature for the smectic polymers was possible using SAXS measurements. A particular arrangement in one of the studied SCLCPs is related to the coexistence of two different phases.
